How is the thermal denaturation of protein determined spectrophotometrically?  

The thermal transition for a sample of chymotrypsinogen has been determined using UV Vis spectroscopy and the CuveT peltier thermostat by analyzing the absorbance over the temperature range between 20ºC to 85ºC. The reversible process of protein denaturation with two distinguishable states corresponding to the native and denatured with respect to temperature is confirmed.
